MATERIALS AND METHODS FOR TESTING NORMAL PHYSIOLOGY OF BLOOD CELLS


3.1 Research Design:  The study is an  experimental, prospective and descriptive  research design.
3.2  area of study:  The study  area is  Nnewi metropolis in  Anambra State, is  a cosmopolitan town.  It  has a population of   4,177,828 according  to  2006 census  (national  bureau of  statistics; 2009)
3.3. Study population: The  population is motorcyclists aged  21-50  and apparently healthy
students of the same   age bracket  that  serves as the control subjects.
3.4  sample size:  50  motorcycle riders,  20  control subjects of age and sex   matched  who are apparently healthy.
3.5  instrumetns for samples collection :  2mls syringers sna dneedles, dry  cotton  woll, spirit swab, EDTA –containers, EDTA Container stand
3.6  method of samples collection:  2mls of blood was collected into EDTA –Bottles   through venepuncture. They were all analyzed immediately after collection, but  where immediate analysis was not possible the sample  were  stored at  4oC for  24   hours
3.7  statistical analysis:  for statistical analysis of the samples  the  mean,  standard deviation, and analysis  of variance  (ANOVA) was used.

3.8  Materials used:
1.      diluted cell suspension
2.      diluents (drabkin’s solution, trisodium citrate, ammonium  oxalate).
3.      Sysmex  machine
4.      Clean grease –free slides
5.      Spreader
6.      Cotton wool.
7.      EDTA –anti –coagulated blood
8.      Leishmann stain
9.      Methanol
10. Buffered water
11. Erythrocyte sedimentation rate (ESR)  TUBES AND  STANDS.
12. Pipette filler
13. Stop watch
14. Capillary  tubes
15. Micro-haematocrit reader and centrifuge
16. Spectrophotometer
17. Microscope
18. Improved neubauer conting chamber (INCC)
19. Counting  chamber cover galss
20. A safe filling  device
21. Hand counter
